How Does PDGF Work? Mechanism of Action, Per the Research
Platelet-derived growth factor (PDGF) is a family of dimeric proteins that bind two receptor tyrosine kinases, PDGFRα and PDGFRβ. Receptor dimerisation triggers autophosphorylation and recruitment of signalling proteins including PI3K, PLCγ, SHP-2 and Ras-MAPK components. Published reviews describe downstream effects on mesenchymal cell proliferation, migration and survival, and on pericyte and fibroblast biology. Much mechanistic detail comes from cell culture and animal models. This page summarises that literature and is educational only.
Platelet-derived growth factor (PDGF) is one of the best-characterised growth factor families in mammalian biology. This page summarises how published pharmacology and review literature describe its receptor targets, intracellular signalling and downstream tissue effects. What PDGF Is, According to the Literature
A 2008 review in Genes & Development described PDGF as a family of disulphide-bonded dimeric molecules assembled from four polypeptide chains, A, B, C and D, producing the homodimers PDGF-AA, -BB, -CC and -DD plus the heterodimer PDGF-AB, and reported that these ligands act principally on cells of mesenchymal origin (PMID 18483217). The same review reported that PDGF was originally identified as a serum constituent released from platelet alpha granules, which is the origin of the name (PMID 18483217).
A 2025 review of PDGF/PDGFR signalling across organ systems reported that the pathway participates in development, tissue homeostasis and repair in multiple tissues, and that its dysregulation has been linked to fibrotic and proliferative disease states (PMID 39482238).
Receptor Targets: PDGFRα and PDGFRβ
The 2008 Genes & Development review reported that PDGF ligands signal through two structurally related receptor tyrosine kinases, PDGFRα and PDGFRβ, which contain five extracellular immunoglobulin-like domains and an intracellular split tyrosine kinase domain (PMID 18483217). Because the ligands are dimers, a single ligand molecule can engage two receptor chains simultaneously, and the review reported that this produces αα, αβ and ββ receptor dimers depending on which ligand is present (PMID 18483217).
A 2018 review in Molecular Aspects of Medicine described the PDGF/PDGFR axis as a validated drug target and reported that several approved tyrosine kinase inhibitors act in part by blocking PDGFR kinase activity (PMID 29137923). That receptor-level pharmacology is one reason the pathway has been mapped in unusual detail.
Ligand-receptor pairing as described in reviews
| Ligand | Receptor dimers described | Source |
|---|---|---|
| PDGF-AA | αα | PMID 18483217 |
| PDGF-AB | αα, αβ | PMID 18483217 |
| PDGF-BB | αα, αβ, ββ | PMID 18483217 |
| PDGF-CC | αα, αβ | PMID 18483217 |
| PDGF-DD | ββ | PMID 18483217 |
PDGF-BB is the most promiscuous ligand in this scheme, which is partly why it appears so often in experimental work; the 2008 review reported that PDGF-BB can engage all three receptor dimer configurations (PMID 18483217).

Try it freeThe Established Signalling Mechanism
The core mechanism is standard receptor tyrosine kinase biology, and it is the part of the picture that reviews treat as settled rather than proposed.
- Ligand binding and dimerisation. The 2008 review reported that dimeric PDGF cross-links two receptor molecules, bringing their intracellular kinase domains into proximity (PMID 18483217).
- Autophosphorylation. The same review reported that receptor dimerisation leads to trans-autophosphorylation of tyrosine residues in the intracellular domain, both activating the kinase and creating docking sites (PMID 18483217).
- SH2-domain recruitment. Phosphotyrosines recruit SH2-domain-containing effectors; the review described PI3-kinase, phospholipase C-γ, Src family kinases, SHP-2 and adaptor proteins such as Grb2 among the partners documented for PDGFRs (PMID 18483217).
- Downstream cascades. A 2022 review reported that PDGFR activation feeds the Ras/MAPK, PI3K/AKT and PLCγ cascades, which together influence proliferation, survival, cytoskeletal rearrangement and migration (PMID 35074329).
- Termination. The 2008 review reported that signalling is attenuated by receptor internalisation and degradation and by phosphatase activity (PMID 18483217).
Reviews consistently describe PDGF as acting over short distances: the 2008 review reported that PDGF-AA and PDGF-BB contain retention motifs that bind extracellular matrix, limiting diffusion and creating local gradients around producing cells (PMID 18483217). That matrix retention is central to how the pathway is understood to work as a paracrine, rather than endocrine, signal.
Downstream Physiology: What Cells Respond
Pericytes and vascular mural cells
The 2008 review reported that PDGF-B signalling through PDGFRβ is required for recruitment of pericytes and vascular smooth muscle cells to developing vessels, a role established through genetic models in mice (PMID 18483217). A 2020 study in JCI Insight reported that elevated PDGF-BB in subchondral bone stimulated angiogenesis and contributed to osteoarthritis development in an animal model (PMID 32208385), animal work, and labelled as such.
Fibroblasts and fibrosis
A 2016 review of the PDGF system in liver fibrosis reported that PDGF isoforms act as potent mitogens for hepatic stellate cells and that PDGFRβ signalling is a driver of the fibrogenic response, with antagonist strategies examined experimentally (PMID 26547628). The 2025 organ-systems review similarly reported fibroblast activation as a recurring downstream consequence of PDGFR signalling across tissues (PMID 39482238).
Pulmonary vasculature
A 2005 study in the Journal of Clinical Investigation reported that PDGF signalling was implicated in pulmonary arterial hypertension, with receptor blockade examined as an experimental approach to vascular remodelling in that setting (PMID 16200204).
Cardiac tissue
A 2019 Nature study reported that activation of the PDGF pathway linked LMNA mutation to dilated cardiomyopathy, identifying PDGF signalling as a mechanistic intermediate in a genetic cardiac model (PMID 31316208).
Bone and repair models
A 2023 study in npj Regenerative Medicine reported that PDGF inhibited BMP2-induced bone healing in an experimental model, which is a useful counterexample to the assumption that a growth factor is uniformly pro-regenerative (PMID 36631491). Context determines direction of effect in this literature.

Get the appEstablished Mechanism vs. Proposed Mechanism
It is worth separating the two tiers explicitly.
| Tier | What the literature describes |
|---|---|
| Established molecular mechanism | Dimeric ligand binding, PDGFRα/β dimerisation, autophosphorylation, SH2 effector recruitment, and activation of PI3K/AKT, Ras/MAPK and PLCγ cascades, as described in reviews (PMID 18483217, PMID 35074329). |
| Established developmental role | Mural cell recruitment and mesenchymal cell development demonstrated through genetic models (PMID 18483217). |
| Proposed / under investigation | Stromal PDGFR signalling as a modifiable contributor to tumour behaviour (PMID 38980580); PDGF-driven angiogenesis as a contributor to joint pathology in animals (PMID 32208385); neuronal-receptor coupling in brain tumour growth (PMID 40897174). |
PDGF Signalling in Tumour Stroma
A 2024 review in Cancer Metastasis Reviews reported that PDGF receptors expressed on stromal cells, rather than on tumour cells themselves, carry functional and clinical significance in tumour biology (PMID 38980580). A 2024 study in Cell Reports Medicine reported that targeting PDGF signalling in cancer-associated fibroblasts blocked feedback activation of HIF-1α and reduced tumour progression in clear cell ovarian cancer models (PMID 38670097), preclinical model work.
A 2025 study in Neuron reported aberrant coupling of glutamate receptors and tyrosine kinase receptors as a mechanism allowing neuronal activity to influence brain-tumour growth (PMID 40897174). This illustrates that receptor tyrosine kinase mechanisms continue to be extended by new experimental findings rather than being fully closed.

Start learning freeWhy Mechanism Does Not Equal Outcome
Several themes in this literature caution against reading a signalling diagram as a prediction of effect. The 2023 bone study reported inhibition, not enhancement, of BMP2-induced healing (PMID 36631491). The 2020 osteoarthritis work reported that elevated PDGF-BB contributed to disease development in an animal model (PMID 32208385). Meanwhile much clinical interest in the pathway has concerned blocking it: a 2018 review described PDGF/PDGFR as a drug target for inhibition (PMID 29137923), and a 2022 review likewise reviewed inhibitor strategies in oncology (PMID 35074329).
PDGF Pathway Signalling: What Studies Report
Because the verified literature here is largely mechanistic and preclinical, it describes pathway consequences rather than a catalogue of human adverse events. Researchers reported that excessive PDGF pathway activity has been associated with fibrogenesis in liver models (PMID 26547628), with vascular remodelling in pulmonary arterial hypertension (PMID 16200204), and with cardiomyopathy in an LMNA-mutation model (PMID 31316208). The 2025 cross-organ review reported that dysregulated PDGF/PDGFR signalling features in pathology across several organ systems (PMID 39482238). No dosing information appears in this page because the verified sources here characterise mechanism, not administration regimens.

Try it freeOpen Questions in the Mechanistic Literature
- Isoform specificity. The 2008 review reported distinct biological roles for different ligand-receptor combinations, but much experimental work still uses PDGF-BB as a general stimulus (PMID 18483217).
- Cell-type context. The 2024 stromal review reported that receptor function depends heavily on which stromal compartment expresses it (PMID 38980580).
- Feedback loops. The 2024 ovarian cancer study reported feedback activation of HIF-1α downstream of PDGF signalling in fibroblasts (PMID 38670097).
- Translation gap. The 2018 drug-target review reported that despite clear molecular rationale, translating PDGFR targeting into clinical benefit has been uneven (PMID 29137923).
Summary of the Mechanism
In the terms used by published reviews, PDGF works by cross-linking two PDGFRα or PDGFRβ molecules with a dimeric ligand, triggering kinase activation and autophosphorylation, recruiting SH2-domain effectors, and switching on PI3K/AKT, Ras/MAPK and PLCγ cascades that drive proliferation, migration and survival in mesenchymal cell types (PMID 18483217, PMID 35074329). Everything beyond that, which tissues, in what direction, under what conditions, depends on the model system, as the organ-by-organ review reported (PMID 39482238). Frequently asked questions
What receptors does PDGF bind?▾
Published reviews reported that PDGF ligands bind two receptor tyrosine kinases, PDGFRα and PDGFRβ, each with five extracellular immunoglobulin-like domains and a split intracellular kinase domain (PMID 18483217). Because PDGF ligands are dimers, they cross-link two receptor chains, producing αα, αβ or ββ receptor dimers depending on the ligand isoform involved (PMID 18483217).
What signalling pathways does PDGF activate?▾
Researchers reported that receptor autophosphorylation creates docking sites for SH2-domain proteins including PI3-kinase, phospholipase C-γ, Src family kinases and SHP-2 (PMID 18483217). A 2022 review reported that these interactions feed the Ras/MAPK, PI3K/AKT and PLCγ cascades, which influence proliferation, survival, cytoskeletal remodelling and cell migration (PMID 35074329).
Which cells respond to PDGF?▾
Reviews reported that PDGF acts principally on mesenchymal cells, including fibroblasts, smooth muscle cells and pericytes (PMID 18483217). A 2016 review reported that hepatic stellate cells respond strongly to PDGF isoforms in liver fibrosis models (PMID 26547628), and a 2025 review reported fibroblast activation as a recurring downstream consequence across multiple organ systems (PMID 39482238).
Is PDGF always pro-healing?▾
No. The literature reports context-dependent effects. A 2023 study reported that PDGF inhibited BMP2-induced bone healing in an experimental model (PMID 36631491), and a 2020 animal study reported that elevated PDGF-BB in subchondral bone stimulated angiogenesis and contributed to osteoarthritis development (PMID 32208385). Direction of effect varied with tissue and model.
Why is the PDGF pathway studied as a drug target?▾
A 2018 review described the PDGF/PDGFR pathway as a drug target and reported that several tyrosine kinase inhibitors act partly by blocking PDGFR kinase activity (PMID 29137923). A 2024 review reported that stromal PDGF receptors carry functional and clinical significance in tumour biology, making the stromal compartment a focus of investigation (PMID 38980580).
What is established versus proposed in PDGF mechanism?▾
Established elements include dimeric ligand binding, receptor dimerisation, autophosphorylation and downstream PI3K/AKT, Ras/MAPK and PLCγ signalling (PMID 18483217). Proposed or still-investigational elements include PDGF-driven angiogenesis in joint pathology (PMID 32208385), feedback activation of HIF-1α in cancer-associated fibroblasts (PMID 38670097), and neuronal-receptor coupling in brain tumours (PMID 40897174).
Does PDGF act locally or throughout the body?▾
A 2008 review reported that PDGF-AA and PDGF-BB contain retention motifs that bind extracellular matrix, limiting diffusion and creating local concentration gradients around producing cells (PMID 18483217). On that basis the pathway is generally described as paracrine or autocrine signalling near the source, rather than a systemic endocrine signal.
