Prodynorphin (abbreviated PDYN, and sometimes written as preprodynorphin) is a precursor protein produced by cells in the nervous system that is enzymatically cut into a family of smaller opioid peptides, including dynorphin A, dynorphin B and alpha-neoendorphin. It is one of three classical opioid peptide precursors in mammals, alongside proenkephalin (PENK) and proopiomelanocortin (POMC). What Class of Molecule Is It?
Prodynorphin is a polypeptide precursor — a longer chain that carries several shorter peptide sequences within it. After the precursor is produced, processing enzymes cleave it at specific sites to release the mature dynorphin peptides, which act primarily at the kappa-opioid receptor. Because the precursor is the upstream source of all of these peptides, measuring prodynorphin or its derived fragments is one way investigators estimate the activity of this signalling system.
Where It Comes From
Prodynorphin is endogenous: it is made by the body rather than obtained from an outside source. Expression is concentrated in neurons of the brain and spinal cord, including regions involved in feeding, stress, defensive behaviour, reward and olfaction. Investigators most often detect it by measuring Pdyn messenger RNA inside neurons, by using genetically modified animals in which prodynorphin-expressing cells can be visualised or manipulated, or by immunoassay of prodynorphin-derived peptides in cerebrospinal fluid, serum or tissue.
How the Term Is Used in Peptide Research
Three usages dominate the published literature:
- As a cell-type marker. Papers describe "prodynorphin neurons" in a given brain region and then map where those cells send and receive signals.
- As a gene under study. Human genetics papers examine variants (polymorphisms) or DNA methylation of PDYN and relate them to behavioural or clinical measures.
- As a measurable peptide. Clinical studies quantify prodynorphin-derived peptides in cerebrospinal fluid or serum and compare groups.

Circuit mapping in animals
A 2026 Neuron paper described a dorsal hippocampus to prodynorphinergic dorsolateral septum to lateral hypothalamus circuit that mediated contextual gating of feeding in mice (PMID 41687615), and the same work was released earlier as a 2024 preprint reporting that circuit (PMID 39149322). In a separate 2025 report, researchers reported that prodynorphin-expressing neurons in the ventromedial hypothalamus responded to a predatory odour and modulated defensive behaviours (PMID 40582282). A 2021 tract-tracing study in The Journal of Comparative Neurology characterised the input and output connections of prodynorphin neurons in a pontine region designated LJA5 (PMID 32602558).
Other anatomical work has catalogued where these cells sit. A 2022 study in Frontiers in Neural Circuits profiled prodynorphin and preproenkephalin mRNA-expressing neurons in the anterior olfactory tubercle of mice (PMID 35937204). Behaviourally, a 2009 study in Neuropsychopharmacology reported that prodynorphin-derived peptides acted as modulators of anxiety and regulated neurochemistry and corticosterone (PMID 18800067).
Human fluid measurements
Because prodynorphin-derived peptides can be assayed in cerebrospinal fluid, several clinical groups have compared patient and control samples. A 2021 study in Movement Disorders reported that cerebrospinal fluid levels of prodynorphin-derived peptides were decreased in Huntington's disease (PMID 33006791). A 2022 paper in the International Journal of Molecular Sciences examined prodynorphin and proenkephalin in the cerebrospinal fluid of patients with sporadic Creutzfeldt-Jakob disease (PMID 35216166). These were observational comparisons, not treatment studies, and the papers did not establish that prodynorphin caused the conditions studied.
Genetics, epigenetics and substance use
The PDYN gene has been examined repeatedly in substance-use research. A 2017 study in Progress in Neuro-Psychopharmacology & Biological Psychiatry investigated genetic variation and epigenetic modification of the prodynorphin gene in peripheral blood cells in alcoholism (PMID 28336495). A 2022 study reported on PDYN gene polymorphisms in Turkish patients with methamphetamine use disorder, together with changes in serum prodynorphin levels during withdrawal and relationships with temperament and depression measures (PMID 32597371). In animals, a 2016 study in Drug and Alcohol Dependence reported that repeated nicotine exposure modulated prodynorphin and pronociceptin levels in the reward pathway (PMID 27430399).
Cognition has also been studied. A 2015 paper in Frontiers in Behavioral Neuroscience reported that a functional polymorphism in the prodynorphin gene affected cognitive flexibility and brain activation during reversal learning (PMID 26190983).
