Where homotrimers come from
Homotrimers are not a single chemical class. The label applies to any protein or peptide that happens to assemble in threes, so the entries grouped under it come from very different origins:

Oligomeric state and function
Part of why the term is used precisely is that assembly state has been reported to track with behaviour. A 2025 study reported that angiopoietin-like 3 monomers were abundant in human plasma but were unable to inhibit endothelial lipase (PMID 41148215) — an example in which the unassembled form and the assembled form were not functionally interchangeable. Separately, a 2018 study reported that assembly of protein complexes restricted the diffusion of Wnt3a proteins (PMID 30320232), indicating that oligomerisation can also alter how far a secreted protein moves. Both findings are reasons that papers specify homotrimer rather than simply naming the protein.
Limits of the term
Calling something a homotrimer says how many identical chains are present in the resolved or measured assembly. It does not say whether the trimer is obligate or transient, whether it exists at the same proportion in a cell as in a purified sample, or what the complex does. Several proteins exist in equilibrium between monomeric and oligomeric forms, and the fraction in each state can depend on concentration, buffer, temperature and post-translational modification. Structural methods such as cryo-electron microscopy and X-ray crystallography report the state captured under the conditions used, which is why papers describe sample preparation alongside the architecture. Homotrimer is therefore a structural descriptor to be read together with the method and conditions in the source paper, not a standalone claim about activity.
